Saved games are to be loaded from the memory card in exactly the same way as when running an original disc. The fact that you are using a backup has nothing to do with it.

Most games start up with an option to either load a saved game or start a new game, while others require the user to navigate some menus to find the command to load a saved game. I have no idea how it's done with tekken games since they are not my 'flavour' (I'm mainly an RPG player), so you will have to find someone else to tell you that. Or just check it in the game manual that comes with every original game. (Unless the dog ate it... )

    Best regards: dlanor

Tekken 5 will try to detect saves on your memory card on startup. This is also true even if the auto-save feature is disabled. If it finds game save data, then it will load all your saved data. If you want it to auto-save everytime you do something in the game, you have to go into "Options > Memory Card (PS2)" from the main menu and set the "Auto Save" feature to "ON". You can load and save your game save data from there also.
