HDD is not a requirement, except for a few versions that were designed to use only HDD for ROMs. (eg: Kaylakaze's versions of InfoGB and InfoNES).

You dont. Only PS2 programs can be run from memory cards, and imbNES is not a PS2 program. It is a PS1 program. The only way to run it on a PS2 is to make a boot CD for it. Also, since that emulator was designed to read ROMs only from a rombank file on the CD, that is how you must store its ROMs.
There is greater freedom in how you use the emulators that are PS2 programs, though each of those programs may have its own limitations as to where you can place ROMs etc.

Booting any programs directly from memory card is impossible on an unmodded console, but it can be done indirectly by booting either a SwapMagic disc or a PS1 exploit disc, either of which can be used to launch a PS2 program from a memory card.

Most emulators can be run from USB, but that doesn't necessarily mean that they can load ROMs from that drive. The newest version of SNES-station can do it, but most other emulators can't.
As for directly booting any programs from USB, that is another impossibility. But this too can be done indirectly, either through a SwapMagic disc which can launch programs from USB, or through the PS1 exploit which can launch a program from memory card, which can in turn be used to launch the emulator from USB. And with the MI chip you mentioned having in a recent PM, you can also use its Dev1 feature to boot a program from memory card, thus eliminating the need for the PS1 exploit.
