2. Need correct location, correct naming.
Everything explained here:
Official OPLv0.8 User Guides
3. Performance increase is not really a decisive argument to use ISO. I mean you should not convert your old ul games for this only reason.
But however ISO files provide more flexibility, ease of use. You have one single file, you have a clear naming of the game (and in 0.9 we plan to push the game name limit to 64 or 128 characters, here it is only 32 to be compliant with ul format), you don't need the additional process of splitting files, you can burn your ISO if you need a DVD of it, ...
If you want to convert your ul games back to ISO you can use the "opl2iso.exe" that you can find in the guide above.
No parameter to list the game from the ul.cfg file, then "iso2opl SLES_123.45" for every game you want to convert back to ISO

For your last question, google answer:
To convert a volume to NTFS from the command prompt
1. Open Command Prompt. Click Start, point to All Programs, point to Accessories, and then click Command Prompt.
2. In the command prompt window, type: convert drive_letter: /fs:ntfs
For example, typing "convert D: /fs:ntfs" would convert drive D: with the ntfs format. You can convert FAT or FAT32 volumes to NTFS with this command.
