I thought the whole issue with large files was the FAT32 bit, hence the need to HTP, does Iris read NTFS externals ?

Nope, the files still get split as long as you're backing up from the disk or you use split4g on the large files. Iris will detect that there's split files and use the internal cache spot of the PS3 to join the file back together and make the game playable. Essentially, it's a workaround to the 4gb limitation.
